Background Information:

The InCELL Hunter Human GLP Stable Cell Line Target Engagement Assay (A549) utilizes the GLP Target Engagement cell line. The stable engineered cell line is intended for use in studying small molecule drug candidates that engage with regulators of epigenetic pathways. This cell line enables assessment of compound-engagement-based stabilization of GLP via detection of GLP levels. This product uses EFC technology. The cell line included overexpresses GLP's methyltransferase domain fused to PL. In the absence of a compound that binds and stabilizes the target, this protein fragment is naturally degraded, and no EFC signal is produced. When the compound engages and stabilizes the target, EA contained in the detection reagent produces EFC signal.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
