Background Information:
The PathHunter Human IKZF3v1 Degradation Stable Cell Line Assay (HEK 293) utilizes the HEK 293 IKZF3v1 Degradation cell line. The stable engineered cell line is designed for use in studying drug candidates (small molecules or biologics) that modulate cereblon E3 ligase affinity. This cell line enables the detection of IKZF3 (isoform 1) levels, allowing for the assessment of targeted proteasome-mediated IKZF3 (isoform 1) degradation. These cells natively express high levels of target E3 ligase cereblon, as well as overexpressing IKZF3 target protein fused to PL. EA is contained in the detection reagent, allowing production of EFC signal proportional to the amount of IKZF3 in the system. Upon proteasome-mediated degradation of IKZF3, the EFC signal is lost.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10 plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
Eurofins DiscoverX optimized Detection Reagents are required for optimal assay performance and must be used with these cell lines in accordance with the cell line license agreed to upon purchase of this cell line assay kit.
