Background Information:

The PathHunter® Human APRIL (BCMA) Signaling Reporter Cell-Based Assay Kit (K562) utilizes the K562 BCMA Signaling Pathway Reporter cell line. This engineered cell line monitors the activation of specific signaling pathways, utilizing the established PathHunter Enzyme Fragment Complementation (EFC) technology. It is designed for use studying drug candidates that affect the NF-kB pathway. This cell line enables assessment of NF-kB stimulation based on changes in the expression of an NF-kB response element-driven reporter protein. The included cell line overexpresses BCMA, and expresses PL-tagged reporter protein under the control of an NF-kB-inducible transcriptional response element. Pathway activation induces expression of the PL-tagged protein, allowing detection of the chemiluminescent EFC signal upon addition of the EA-containing detection reagent.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
