Background Information:

The PathHunter® Human TREM1-DAP12 Signaling Reporter Cell-Based Assay Kit (Jurkat) utilizes the Jurkat TREM1-DAP12 Signaling Pathway Reporter cell line. This engineered cell line monitors the activation of specific signaling pathways, utilizing the established PathHunter Enzyme Fragment Complementation (EFC) technology. It can be used for studying drug candidates (small molecules or biologics) affecting the TREM1 pathway. This cell line enables assessment of TREM1 pathway stimulation based on changes in the expression of a NFAT response element-driven reporter protein. The included cell line overexpresses TREM1 and DAP12, and also expresses PL-tagged reporter protein under the control of a NFAT-inducible transcriptional response element. Pathway activation induces expression of the PL-tagged protein, allowing detection of the EFC signal upon addition of the EA-containing detection reagent.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
