Background Information:
The PathHunter® Human OPRK1 (Opioid Kappa) β-Arrestin Cell-Based Assay Kit (U2OS) utilizes the Human OPRK1 β-Arrestin cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of β-arrestin recruitment following receptor activation for GPCR drug discovery and development programs. This cell line enables the assessment of ligand (e.g., dynorphin A) based activation of OPRK1 GPCR activity via detection of β-arrestin recruitment. This cell line employs Enzyme Fragment Complementation (EFC) technology that combines an overexpressed PK-tagged OPRK1 target receptor and an EA-tagged β-Arrestin 2 to generate a chemiluminescent signal proportional to receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
