InCELL Hunter™ Human BCL2L1-BAX Stable Cell Line Target Engagement Assay (U2OS)

Background Information:

The InCELL Hunter Human BCL2L1-BAX Stable Cell Line Target Engagement Assay (U2OS) utilizes the BCL2L1-BAX Target Engagement Cell Line. This stable engineered cell line provides an easy-to-use cell based assay to study small molecule drug candidates that disrupt protein binding. This kit assesses compound-based disruption of BCL2L1-BAX binding via detection of BAX levels. This product uses EFC technology. The cell line included overexpresses BXL2L1 target in an untagged form and BAX binding partner fused to PL. When a compound engages the BCL2L1 target protein, partner binding is disrupted and the destabilized BAX is degraded, leading to a loss of EFC signal upon addition of EA-containing detection reagent.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay. Assay plates and positive control for use in the assays can be purchased separately. For continued use of the cell line, additional quantities of cell culture and assay reagents are available for purchase separately.
