Background Information:
The KILR® Jurkat (Human) PD-1 Cytotoxicity Assay (Clone) utilizes the KILR® Jurkat PD-1 cell line. The cell line is a stable, engineered target line for measuring direct cell death mediated by immuno-oncology treatments (e.g., checkpoint inhibitors, checkpoint agonists, bispecifics, CAR-T, and more) via Antibody-Dependent Cell-mediated Cytotoxicity (ADCC), Complement-Dependent Cytotoxicity (CDC), and Antibody-Dependent Phagocytosis (ADCP). This product uses EFC technology. The line overexpresses a ProLabel-tagged housekeeping gene as well as untagged PD-1. Once the cells have been lysed by effector cells, ProLabel is released into the media, where EA, contained in the detection kit, produces an EFC signal proportional to the level of ProLabel released. This allows for the calculation of the percent lysis of the line.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
