Background Information:

The PathHunter® Human MCP-4 (CCR3) β-Arrestin Cell-Based Assay Kit (CHO-K1) utilizes the Human CCR3 β-Arrestin cell line. This engineered stable cell line is optimized for GPCR pharmacology and mechanism-of-action (MOA) studies, enabling quantitative assessment of β-arrestin recruitment following receptor activation for GPCR drug discovery and development programs. This cell line enables the assessment of ligand (e.g., CCL13/MCP-4) based activation of CCR3 GPCR activity via detection of β-arrestin recruitment. This cell line employs Enzyme Fragment Complementation (EFC) technology that combines an overexpressed PK-tagged CCR3 target receptor and an EA-tagged β-Arrestin 2 to generate a chemiluminescent signal proportional to receptor activation. This product leverages a sensitive chemiluminescent detection system that delivers a wide dynamic range and robust Z’ for potency, efficacy, and rank‑order profiling across small molecules and biologics.
This kit includes a stable engineered cell line (supplied as two vials of cryopreserved cells); cell line specific reagents for cell thawing, propagation, detachment, cryopreservation, and cell plating; and a 10‑plate detection kit required to run the assay.
